PENGEMBANGAN SIDIK JARI DNA KLON KARET IRR SERI 400 BERDASARKAN MARKA SSR

Pengembangan areal perkebunan karet ke daerah non tradisional merupakan tantangan bagi pemulia untuk menghasilkan klon-klon karet unggul baru berproduksi tinggi dan sesuai dengan kondisi agroklimat tertentu. IRR seri 400 merupakan salah satu seri klon karet unggul baru yang sedang dalam tahap uji adaptasi dan persiapan pelepasan ke masyarakat. Dalam rangka persiapan dan penyebaran klon karet unggul baru tersebut diperlukan penyiapan kebun entres yang murni secara morfologi dan molekuler. Pengembangan metode identifikasi klon karet secara akurat melalui molekuler perlu dilakukan untuk menjaga kemurnian kebun entres dalam upaya menjaga mutu genetik dari suatu klon. Penelitian ini bertujuan untuk menganalisis sidik jari DNA 10 klon karet IRR seri 400 sebagai penanda genetik setiap klon menggunakan 10 primer SSR (Simple Sequence Repeat). Hasil amplifikasi PCR 10 lokus SSR pada 10 klon menghasilkan 53 alel dengan rata-rata 5,3 alel per lokus. Berdasarkan analisis visual skoring posisi alel masing-masing lokus berhasil diidentifikasi 33 marka spesifik klon dengan 2-6 marka pada masing-masing klon. Marka spesifik klon tersebut dapat digunakan sebagai set marka DNA penciri untuk identifikasi 10 klon IRR seri 400 serta pengembangan database yang berguna untuk pengelolaan dan perlindungan plasma nutfah karet Indonesia dari pencurian dan klaim pihak asing.
